Factors affecting Discontinuation of Paliperidone Long Acting Injection

Abstract
Objectives£ºPaliperidone palmitate long-acting injection(PP) is an effective and broadly well tolerated treatment in practice. The aim of this study was to determine factors relating to discontinuation with treatment with PP, we analyzed the time course and causes of PP discontinuation that occurred in 12 months period in a clinical setting.

Methods£ºRetrospective database of 81 patients with schizophrenia were reviewed for analyzing the duration of PP and the reasons for discontinuation(from June 2011 to November 2016), with the motives for discontinuation coded. The causes for termination were analyzed.

Results£ºOverall, 58% patients discontinued PP beyond 12 months from initiation. The common reasons for discontinuation were follow-up loss(42.5%), no effect(19.1%), side effect(19.1%). extrapyramidal symptom accounted for 44.4% of all side effect inducing discontinuation. Pain, somnolence accounted for 22.2% of side effect, respectively. Inpatient initiation, switched from risperidone, risperidone long-acting injection(LAI), compliance before injection, hospitalization during treatment course, GAF score at last LAI, CGI-S at last LAI were associated with discontinuation on univariate analysis. However, only hospitalization during treatment course was independently associated with discontinuation on multivariate analysis.

Conclusion£ºHospitalization after PP was associated with 12-month PP discontinuation. Prospective researches using ore objective tools are needed to clarify the reasons for PP discontinuation.
